肉毒桿菌神經(jīng)毒素（BoNT）是人類已知的毒性大的物質(zhì)之一。已經(jīng)鑒定了七種免疫學(xué)上不同的神經(jīng)毒素血清型，命名為A至G型（BoNT / A，BoNT / B，BoNT / C，BoNT / D，BoNT / E，BoNT / F，BoNT / G）。這些肉毒桿菌神經(jīng)毒素天然產(chǎn)生與一種或多種無(wú)毒神經(jīng)毒素相關(guān)蛋白（NAP）復(fù)合。NAP對(duì)于攝入毒性很重要，因?yàn)樗鼈儽Ｗo(hù)神經(jīng)毒素免受蛋白酶，酸性，熱的影響，并且它們也可能在粘膜層的易位中發(fā)揮作用。
合成為單個(gè)150kDa多肽鏈，隨后通過(guò)切割激活復(fù)合物的實(shí)際神經(jīng)毒素部分以產(chǎn)生兩條鏈，重鏈和輕鏈，其通過(guò)單個(gè)二硫鍵連接。對(duì)于每種毒素，50kDa輕鏈?zhǔn)卿\??依賴性蛋白酶，其切割神經(jīng)傳遞期間突觸小泡膜融合所必需的單個(gè)靶蛋白。A，C和E型神經(jīng)毒素特異性結(jié)合并選擇性地切割突觸體相關(guān)蛋白SNAP-25，而B(niǎo)，D，F(xiàn)和G型切割突觸小泡蛋白-2，也稱為VAMP-2。靶蛋白的切割抑制神經(jīng)元之間的神經(jīng)遞質(zhì)釋放，這導(dǎo)致肌肉麻痹。

每種毒素很容易分為三個(gè)區(qū)域，反映出對(duì)毒性很重要的三個(gè)功能; 特異性底物的結(jié)合，易位和切割。重鏈負(fù)責(zé)識(shí)別突觸前膜上的特定神經(jīng)節(jié)苷脂和特定蛋白質(zhì)受體。重鏈的易位結(jié)構(gòu)域部分介導(dǎo)輕鏈穿過(guò)內(nèi)體膜，后靶蛋白被酶促結(jié)構(gòu)域切割。

肉毒桿菌神經(jīng)毒素是研究中有價(jià)值的研究工具，旨在闡明囊泡運(yùn)輸和毒性所涉及的機(jī)制，以及了解突觸傳遞的潛在事件。除了使用150kD全毒素的研究之外，重組無(wú)毒重鏈可用于評(píng)估毒素的結(jié)合特性，并且重組無(wú)毒輕鏈可用于篩選抑制蛋白酶結(jié)構(gòu)域的酶活性。
